A rain collector barrel can supply water to up to 8 plumbable entities (such as sinks, showers, baths) on the floor below it providing they are situated directly under the tiles around the water barrel. Anything placed directly underneath the barrel will not be plumbable. Note that the room in which the plumbable entity is placed must be fully enclosed with walls before the option to plumb becomes available, i.e. you can't just build a "roof" above a sink and place barrels on it.

Before build 41 the Wrench was needed instead of the pipe wrench for Plumbing a sink, which effectively rendered the pipe wrench redundant. However, this was resolved in build 41.
